The xpath that you are using is incorrect. I am providing you the correct xpath and an alternate way to click on the auto-completer (by using className), you can use either of them, both will work fine. And as the wait.until method returns the element, you can directly perform the click on it, which would result in one atleast less operation/scraping on the page.
Autocompleter Correct Xpath:
wait.until(ExpectedConditions.visibilityOfElementLocated(By.xpath("//a[#class='uiSelected']"))).click();
Autocompleter by className:
wait.until(ExpectedConditions.visibilityOfElementLocated(By.className("uiSelected"))).click();

I have searched but havent found something similar to what Im trying to do. I'm using java by the way, I'm trying to click/access an a tag with selenium. The issue is that I'm not sure how to go about it. There seems to be a function/event that I need to set off but not quite sure how to. I tried a few ways as get text and clicking but I knew that wasn't going to work. Also I seen there are ways of using JavascriptExecutor but not sure how to use it for my case. I will post the tag below and alsothe function signature, that might help. If theres a similar question please post the link.

You don't need any JS. Just use this xpath:

You just use cssSelector like below:

Using firefox and marking a link in my web-app I get among other things, this code which I think I can use to caprture an object:

This string is "classname" in Firebug.
Going to the script I type in:

But the script does not find the element when executing.

You cannot use search by compound class name (name with spaces). Try to use search by CSS selector instead:
WebElement rolle = driver.findElement(By.cssSelector(".cb_or_somename_someothername.cb_area_0219"));
or by XPath:
WebElement rolle = driver.findElement(By.xpath("//*[#class='cb_or_somename_someothername cb_area_0219']"));
Also you still can use search by one of two class names:
WebElement rolle = driver.findElement(By.className("cb_or_somename_someothername"));
or
WebElement rolle = driver.findElement(By.className("cb_area_0219")); // Note that this class name could be generated dynamically, so each time it could has different decimal part

If you get Element is not clickable... exception it seem that your element is covered by another element at the moment you try to click on it. So try to wait until this "cover" is no more visible:

Seems that you try to interact with object inside a <svg> element. If so, you cannot manage it's child elements simply using click() method.
Try this instead:

I am new to selenium. I am practicing to write a test case on http://www.countdown.tfl.gov.uk. Below are the steps I followed:
a) I opened the browser to selenium Web Driver
b) Found the search text Box and enter H32 and clicked on search button to selenium.
Till this part it works fine.
Now on the page I am actually getting two records on the left side of the page under the search. I am actually trying to click on the first one i.e. "Towards Southall,Townhall" link. Nothing is happening.

Since you are getting NoSuchElement Exception now, you may try the following code with the usage of WebDriver explicit wait.
WebDriverWait wait = new WebDriverWait(driver, 15);

towardssouthallLink.click();
Or WebDriver implicit wait

Tips:
The search results need some time to be retrieved, so please use Explicit wait or Implicit wait.
Don't use locators like span[#id='ext-gen165'], they are ExtJs auto generated.
In this case, css selector can also be used: #route-search li:nth-of-type(1) > span
You aren't calling selectrouteDirection.
You probably want:

It's already a WebElement anyway.
I found that id for those links are dynamically generated. ids are of the form 'ext-genXXX' where XXX is number which is dynamically generated hence varies each time.
Actually, you should try with linkText:
For 'Towards Southall, Town Hall'
driver.findElement(By.linkText("Towards Southall, Town Hall")).click
For 'Towards Hounslow, Bus Station'
driver.findElement(By.linkText("Towards Hounslow, Bus Station")).click
